Platycodaon grandiflorum in Traditional Chinese Medicine

Collection : The drug is collected in spring and autumn, wash clean, removed from rootlet, peeled when fresh or unpeeled and dried.
Description : Cylindrical or slightly fusiform, gradually becoming tapering downwards, some branched, slightly twisted. 7 - 20 cm long, 0.7 - 2 cm in diameter. Externally white or pale yellowish white or yellowish brown to greyish brown when unpeeled. Longitudinally twisted-furrowed with transverse lenticel-like scars and branched root scars and with transverse striations at the upper part. Sometimes the apex showing in relatively short or inconspicuous rhizome which is marked by several crescent shape stem scars. Texture fragile, fracture uneven, cambium ring brown. Bark whitish with cleft, pale yellowish white. Odourless, taste sweet and then bitter.
Identification : 1.Transverse section: Cork cells remaining occasionally showing cork layers when unpeeled. Cork cells containing small prisms of calcium oxalate. Cortex narrow, often with clefts. Phloem scattered with laticiferous tube groups, walls somewhat thickened containing fine granular yellowish brown contents. Cambium in a ring. Xylem vessels singly scattered or aggregated in groups of several vessels, arranged radiately. Parenchymatous cells containing inulin.2.Mount the section of dried drug with dilute glycerin, examine microscopically. Fan-shaped or subrounded crystals of inulin are visible.Assay: Weigh accurately about 4 g of the powder to a Soxhlet's extractor, macerate in 25 ml methanol for 15 hours, add further 25 ml of methanol. Heat under reflux for 6 hours and allow to stand for overnight. Filter, evaporate the filtrate on a water bathe to 15 - 20 ml, cool. Add 50 ml of ether, shake and allow to stand. Discard the supernatant solution and dissolve the precipitate on heating with quantities of 20, 10 and 5 ml of methanol successively, cool and filter. Combine the methanol solutions, concentrate on a water bath to 15 - 20ml and cool. Add 50 ml of ether, shake then treat in the same manner. Combine the methanol solution, transfer to a tared evaporating dish. Evaporate the solution to dryness on a water bath, dry at 105ºC to constant weight. Calculate the content of total Platycodon saponins. It contains not less than 6.0% of total Platycodon saponins.
Processing : Eliminate foreign matter, wash clean, soften thoroughly, cut into thick slices and dry.
Action : To relieve cough, to sothe sore throat, and to promote expectorationand discharge of pus.
Indication : cough with much phlegm, sore throat, hoarseness; lung abscess with purulent expectoration; abscess that does not open after suppuration
Dosage : 3 to 9 g.
Storage : Preserve in a dry place, protected from mould and moth.
