Ligusticum chuanxiong in Traditional Chinese Medicine

Collection : The drug is collected in summer when the node of stem becomes obviously swollen and purplish, removed from soil, baked gently to dryness after sundried and removed from rootlet.
Description : In irregualar knotty and fish like masses, 2 - 7 cm in diameter. Externally yellowish brown, rough and shrunken with many parallel and raised annulations showing dent and subrounded stem scars on the summit and numerous tuberculous rootlet scars beneath the summit and at the annulations., texture compact, uneasily broken, fracture yellowish white or greyish yellow, scattered with yellowish brown oil cavities. Cambium in an andulate ring. Odour strongly aromatic, taste bitter, pungent and slight numb but sweetish afterwards.
Identification : 1.Transverse section: Cork consisting of over 10 layer of cells. Cortex narrow, scattered with root trace vascular bundles, interfascicular camnium distinct. Phloem broad, cambium undulate or irregularly polygonal. Vessels in xylem polygonal or sub rounded, mostly uniseriate or arrange in V shape, wood fibres in bundles occasionally found. Parenchymatous scattered with numerous oil cavities, sub rounded, elliptical or irregular, pale yellowish brown, smaller near the cambium and gradually becoming large outwards. Parenchymatous cells containing abundant starch granules. Some with crystal of calcium oxalate, sub rounded or clustered like.Powder: Pale yellowish brown or greyish brown, starch granules abundant, single granules ellipsoidal oblong, sub spheroidal, ovoid or reniform, 5 - 16 µm in diameter. About 21 µm long, hilum pointed, slit shaped or V shaped. Compoun granules occasionally found, consisting of 2 - 4 components. Crystal of calcium oxalate occurring in parenchymatous cells in sub rounded masses or clustered crystal like, 10 - 25 µm in diameter. Cork cells dark yellowish brown, frequently overlapped in many layers, polygonal in surface view, thin walled. Oil cavities mostly broken, fragments occasionally founnd, secretory cells thin walled containing many oil drops. Vessels mainly spiral and reticulate scalariform as well, 14 - 50 µm in diameter, the thickened walls of some spiral vessels connected to form reticulate like vessels.2.Macerate 1 g of the fine powder in 5 ml of petroleum ether (30 - 60ºC), shake constantly and allow to stand for 10 hours. Evaporate 1 ml of the supernatant to dryness and dissolve the residue in 1 ml of methanol. Add 2 - 3 drops of 2% solution of 3.5-dinitrobenzoic acid methanol and 2 drops of saturated solution of potassium hydroxide in methanol, a purplish red colour is produced.To 1 g of the powder add 20 ml of ether, heat under reflux for 1 hour, filter and evaporate the filtrate to dryness. Dissolve the residue in 2 ml of ethyl acetate as the test solution. Prepare a solution of Rhizoma Chuanxiong reference drug in the same manner as the reference drug solution. Carry out the method for thin layer chromatography (Appendix Vl B), using silica gel G as the coating substance and n-hexane-ethyl acetate (9:1) as the mobile phase. Apply separately to the plate 1 - 2 µl of each of the two solutions. After developing and removal of the plate, dry it in the air and examine under ultra violet light (365 nm). The fluorescence spots in the chromatogram obtained with the test solution correspond in position and colour to the spots in the chromatogram obtained with the reference drug solution.Total ash: Not more than 6.0%
Processing : Eliminate foreign matter, grade according to size, soak briefly, wash briefly clean, soften thoroughly, cut into thin slices and dry.
Action : To promote the flow of blood and qi, dispel wind and relieve pain.
Dosage : 3 to 9 g.
Storage : Preserve in a ventilated and dry place, protected from moth.
