Hyoscyamus niger in Traditional Chinese Medicine

Background

Chinese Name (pinyin): Tianxianzi

Chinese Name  :

Common Name  :Henbane Seed

Specific Name  : Semen hyoscyami

Scientific Name:
Collection  : The fruit is collected in summer and autumn when the pericarp turns yellow, expose to the sun and seed is tapped out, removed from pericarp and branch and dried in the sun

Description  : Flattened subreniform or flattened ovoid, about 1 mm in diameter. Externally brownish yellow or greyish yellow with fine reticulated striations and a pointed hilum at somewhat acute end. Section greyish white oily having endesperm, embryo curved. Odorless, taste slightly pungent.

Identification  : 1.To 0.5g of the powder add 0.5ml of concentrated ammonia TS, mix well, add 5ml of chloroform, stopper tightly, shake constantly for 0.5 hour and filter. Evaporate the filtrate to dryness, dissolve the residue in 0.5ml of chloroform. Evaporate 5 drops of the solution on a water bath to dryness, add 4 drops of fuming nitric acid, evaporate to dryness. To the residue add 1ml of dehydrated ethanol and a small piece of potassium hydroxide, a purple color is produced.2.To 1g of the powder add 10 ml of petroleum ether (30 - 60ºC), ultrasonicate for 15 minutes, discard the petroleum ether solution, treat the residue in the same manner once again. Evaporate the residue to dryness, moisten with 2 ml of a mix solution of equal amount of concentrated ammonia TS and ethanol, add 20 ml of chloroform, ultrasonicate for 15 minutes, filter. Evaporate the filtrate to dryness, dissolve the residue in 0.5ml of dehydrated ethanol as the test solution. Dissolve atropine sulfate CRS and scopolamine hydrobromide CRS in dehydrated ethanol to produce solution containing 1 mg of each per ml as the reference solution. Carry out the method for thin layer chromatography (Appendix Vl B), using silica gel G as the coating substance and ethyl acetate-methanol-concentrated ammonia TS (17:2:1) as the mobile phase. Apply separately to the plate 5 µl of each two solutions. After developing and removal of the plate, dry it in the air. Spray with potassium iodobismuthate TS ethanolic sodium nitrite TS successively. The two brown spots in the chromatogram obtained with the test solution correspond in position and color to the spot in the chromatogram obtained with the reference solution.

Processing  :
