Collection  : The drug is collected in spring and autumn, wash clean and dried.

Description  : Rhizomes irregular, nodular cylindrical, 1.4 - 4.2 cm in diameter. Root cylindrical, mostly tortuous, 3.5 - 12 cm long, 0.3 - 1.5 cm in diameter, external greyish brown or blackish brown, rough with fine longitudinal furrows and wrinkles, bark relatively thin, sometimes exfoliated, the expose surface appearing greyish yellow. Texture hard, fracture yellowish white, fibrous. Odour characteristic and aromatic, taste slightly pungent somewhat bitter and astringent.

Identification  : 1.Transverse section of the root: cork cells of the 10 or more layers. Cortex thin, scattered with secretory canals,most of parenchymatous cells containing cluster of calcium oxalate, 11 - 64µm in diameter. The outside of the phloem scattered with more fibre bundles which lessen inwards, secretory canals subrounded or elliptical, 25 - 51µm long radial, 48 - 97µm long tangentially. Parenchymatous cells containing clusters, cambium in a ring. The majority of the root occupied by xylem, rays 1 - 3 rows of cells wide. Vessels relatively thin walled, mostly several grouped, xylem fibres well develop. Transverse section of the rhizome: Fibre bundles in phloem much more than those in root, pith visible.2.Heat under reflux 5 g of the powder with 20 ml of 75% ethanol for 30 minutes and filter. Evaporate the filtrate to dryness, dissolve the residue in 5 ml of 75% ethanol and filter. To 1 ml of the filtrate and 1m of 3% solution of sodium carbonate and 1 - 2 drops of freshly prepared diazotized á-nitroaniline TS, a red colour is produced. To another 1 ml of the filtrate add 3 - 5 drops of each of 5% sodium nitrite solution and 10% aluminum nitrite solution and 10% aluminum nitrite solution, allow to stand for a moment, then add 2 ml of sodium hydroxide TS, a reddish brown colour is produced. 3.To about 3 g of the powder add 20 ml of 75% ethanol, heat under reflux for 30 minutes, filter and evaporate the filtrate dryness, dissolve the residue with 10 ml of water in tap funnel, extract with 2 quantities each of 5 ml of chloroform. Combine the chloroform solutions, evaporate on a water bath to dryness. Dissolve the residue in 1 ml of methanol as the test solution. Take 5g of Radix Acanthopanacis senticosi reference drug, produced the reference drug solution in the same manner. Take isofraxidin CRS. Add methanol to produce a solution containing 1 mg per ml as the reference solution. Carry out the method for thin layer chromatography (Appendixx Vl B) using silica gel G containing sodium carbonxymethylose as the coating substance and chloroform-methanol (95:5) as the mobile phase. Apply 10µl of each of the three solutions separately to the plate, dry it in the air., examine under ultra violet light (254 nm). The spots in the chromatogram obtained with the test solution correspond in position and colour to the spot in the chromatogram obtained with the drug reference solution. A blue spot in the chromatogram obtained with the test solution correspond in position and color to the spot in the chromatogram obtained with the reference solution.
